Unless she asks you directly if he's cheating I wouldn't volunteer any information. She will find out sooner or later(women always do). I would also let your friend know that you will not lie if asked, and that you don't approve of his behavior.

You should at least talk with your friend about it. In the long run it's his choice what he decides to do about it - tell her, not tell her. As a lot of people say, it's really not your business, but if you're afraid she'll get hurt, saying something to your friend in a calm fashion only advising him against it, couldn't hurt. At least that way you're expressing your concerns for both of them and not just standing idly by while he continues hurting her.. =\
